The 2nd Arc Storyline is unlocked through rebirthing once. It allows you to see new NPCs in old worlds and grants access to The Remnant Zone once you reach Antenna Level 20. Some NPCs, such as Ghost Hunter Archie and Ghost Hunter Cheshire, are part of the 2nd Arc Storyline (as seen in them mentioning the other NPCs), but don't require a rebirth to talk to them due to being unlocked in a different way.
